It's inevitable that the comparison between this game, and the game Rock Band be made. However for me, the original Guitar Hero series is what created this genre and the Harmonix folks took it to the next logical step with Rock Band before they did over at Guitar Hero.
I own the abomination that is Rock Band on the Wii, Guitar Hero III on the Wii, Guitar Hero World Tour and Rock Band 2.
To make it simple, if you own nothing, I would recommend Guitar Hero World Tour over Rock Band 2.
For me, it's about the drum kit. Of the two families I prefer the GH:WT style out of the box. It's three drum/two cymbal approach works the best for me and gives you the most 'real' feel. Yes, I'm aware you can add cymbals to the RB2 family but it's just that - add which makes the system more expensive. Plus, unlike other reviewers, the fact that the GH:WT system uses the Wiimotes as their connection is a huge appeal to me, not a detriment. That makes everything save the microphone a wireless device. With Rock Band the Drums were a tripping nightmare trying to put 'out of the way' and back out when I wanted to play with them. I've had four wiimotes since I bought the system when it came out so this wasn't a 'bad' thing but if you don't have the Wiimotes this can be an issue as you need one per instrument, four total.
The drums aren't perfect on GH:WT though however. Instead of a spot to 'anchor' the foot pedal it floats free. Not being a drummer the foot pedal for me tends to be a stomp pedal and that makes it move around a lot. This is on a carpeted floor.
The new guitar is a bit less boxy on the back side, but in general feels a lot like the GH 3 guitar. I have not used the new touchpad like part as for me sliding up and down the neck I can get lost some times.
The new 'star power' button on the bridge of the guitar controller is a welcome attempt at making it less awkward for those of us who like to sit on our but with our feet up and rock out. However I still get all ham fisted and screw up when I try to punch the power while playing. Glad that the wave around the guitar like a fool option is there as well. It took me some time to figure out (without reading manual) that banging both cymbals is the star power option. I had to pause it and read the manual.
Game Play is now your typical multi-player rock group style game play. No big surprises. I'll admit I do like Rock Band's style of 'save the bad guy' in the group which is important for me as I have a 7 and 5 year old band members sometimes. However I also more like the really 'beginner' mode especially for the guitar. HOld any button down and strum is a great tool for teaching the really little kids and un-coordinated adults how to play and still have fun with everyone else.
However when the five and seven year old go to bed and Dad wants to play the solo mode has one feature I wish they'd spend a little time on. I'd like to have the pre-built characters be selectable as band members and each have some difficulty/expertise etc. Random kick offs of star power, that kind of things. I'm not a big fan of this 'insert some random rock star that was paid a license fee' to enter game and 'battle' that the solo modes have, nor do I care to pretend I'm some rock star.
The online content I've not had a complete time to work with other than buying a few songs I wanted. I have not tried the multiplayer content yet but plan on it.
The GH 3 guitar I had from the first Wii based GH game worked flawlessly with the system.

I really like this game...Have been having a blast playing it. The drums are hard, but I am a bit uncoordinated :) Looking forward to getting a second guitar to play a full band. Also, as the controllers are compatible with rock Band plan on getting that game too.

We had GH3 for the Wii and it was our most played game. The only problem was getting the kids to take turns. Now they don't have to. Our kids (and the neighborhood kids) would play this game everyday if homework, sports and we parents didn't get in the way (we parents play too). The only problem I have with the game is that there should really be a way to block the songs that are inappropriate for kids.
